Production of 186Rhenium- Perrhenate and its using in Preparation of Radiopharmaceutical Compound 186Rhenium–HEDP

The aim of this study was to prepare 186Re solution witch has been used in wet labeling of HEDP (hydroxyethylidene diphosphonate) to obtain 186Re-HEDP complex. Enriched 185Rhenium was irradiated by neutron at a flux of 5×1012 neutron/cm2.s for ten days to produce 186Re. To prepare perrhenate solution some chemical processing like H2O2 oxidation, drying and pH adjustment were carried out. Finally perrhenate solution with specific activity about 350 mci/mgr and radiochemical purity about 99% was obtained.

In preparation of 186Rhenium–HEDP complex different condition for reaction was investigated. Stannous chloride and ascorbic acid were used for reduction of perrhenate. Radiochemical yield was determined by two paper chromatography methods. Preparation of complex was dependent to pH values of reaction mixture, amount of HEDP and SnCl2 concentration.

Under optimal condition the radiochemical yield for 186Rhenium–HEDP reached about 98% and the specific activity was ~ 10 mci/mgr HEDP. Stability of Rhenium-HEDP was studied when stored at 20-25 ºC for 3h, 24h, 48h and 4 days.

Biodistribution studies of 186Re-HEDP in rats showed markedly taken up by the kidneys. 186Re-HEDP had uptake by femur bone (1.01% ID/g) in four hours post injection which remained almost constant up to 24 h. Activity in other organs was negligible after 24 h.
