Solid Forms of Pharmaceutical Molecules (Trospium Chloride).

Screening of solid forms of pharmaceutical molecules is important for drug development. Pharmaceutical molecules can form crystalline and amorphous phases. Crystalline phases include potential polymorphic ansolvates, solvates, salts, solvated salts, cocrystals, solvated cocrystals, cocrystals of salts and solvated cocrystals of salts. Amorphous phases contain amorphous anhydrates and hydrates.

Screening of trospium chloride (antispasmodic drug) was done by crystallization from several solvents and X-ray structures of crystalline products were determined. The structure of disordered anhydrate (polymorph I) was preliminary solved from synchrotron powder diffraction data (group P21, Z = 2) by SA methods in DASH software and refined in Accelrys Reflex. Single crystals of disordered anhydrate (polymorph II) were obtained from ethanol as well. The structure has symmetry P21/c, Z = 4 and was refined by Jana2006 software using rigid body approach. The possibility of multiphase or twinned structures were tested and excluded. During the next crystallization experiments single crystals of methanol and acetone solvates were prepared. Their structures were refined in P21/n, Z = 4.
