A New Strategy for the Synthesis of Nucleosides: One-Pot Enzymatic
Transformation of Pentoses into Nucleosides

We have recently suggested a new strategy for the synthesis of nucleosides
(Bioorg. Med. Chem. 2006, 14, 6327). In the present
communication, preliminary results of one-pot enzymatic transformation
of D-ribose, 2-deoxy-D-ribose, or D-arabinose into the corresponding
pyrimidine and purine nucleosides employing recombinant pure ribokinase
(RK), phosphopentomutase (PPM) and nucleoside phosphorylases (NP),
and the respective pyrimidine or purine heterobases will be reported.
Search for the transformation conditions to achieve an optimal concerted
action of RK [pentose →
pentose-5-phosphate (P-5P)], PPM [P-5P →
pentofuranose-1-a-phosphate (PF-1aP)], and NP (PF-1aP + heterobase
→ nucleoside)
gave rise to the formation of pyrimidine or purine nucleosides in
satisfactory yields.
