Synthesis and analysis of some hydroxypyran -4- one intermediates and their analgesic effects in mice

Introduction
: Hydroxypyran -4- one derivatives have been considered as Iron chelating and suppose to be used in malaria disease.
Recently they have been candidate as analgesic or anti-inflammatory agents' .In this study we decided to synthesize some novel 2-methyl-5-hydroxypyran-4-one as goal compounds. In the way to synthesis final compounds some intermediates were achieved and they were subjected to pharmacological evaluation for their analgesic potentials.

Methods: Kojic acid was used as starting material. It was reacted with thionyl chloride to prepare 2- chloromethy -5-hydroxypyran -4-one. The chloride l group was removed in a reaction used Zn/HCl. New compound was reacted with benzyl chloride in alkaline condition to produce 5-benzyloxy -2 -methyl pyran -4-one.
All compounds and Intermediates were tested after analyzed by IR and NMR as analgesic in mice using formalin test.

Results: The results showed that compounds were synthesized successfully.
Pharmacological data indicated that in addition to kojic acid two of the synthesized derivatives showed analgesic activity in formalin test.

Discussion: Our studies show that 4-hydroxypyran compounds can be synthesized and they may be potentially good analgesic or anti-inflammatory agents. The mechanism of action is not clear yet and further studies are needed.
